Can I use a PCI Express card in a PCI slot?

No theres not enough power or bandwidth in a PCI slot for a PCI Express x16 - plus it wont even fit physically - you need a special dedicated graphics slot on your motherboard

No. It has to be a PCIe slot. The card won't even fit, so you would have found out eventually anyhow =)
