Time from lift off. T minus 2 minutes means 2 minutes until lift off. If the take off is scheduled for 1:00pm and it is 12:58pm (T) then it is T minus 1:00pm which is 2 minutes.

T denotes time in this instant.
Negative T = Time before lift-off
Positive T = Time after lift-off (this counting goes on until this particular mission ends)

Shy Girl is right, and most of the others are wrong.
"T" means "time," as in "It's time, folks, let's do it."
So "T minus 20" means "20 seconds before time to do it."
In this case, "do it" means "energize the squibb," which is the electric igniter that lights the rocket engines.
T does not mean "take off."
