Depends on the fat/muscle ratio of your thighs & butt.
Your muscle size will increase with any sort of consistent exercise. If you have little fat in that area, you may notice an increase in the size of your upper legs and probably calves as well since those are highly engaged in stair climbing.
Doing free squats (just body weight, no added weight) is probably the best way to tone your upper legs and butt.

If you use it in an aerobic manner (lower resistance, many reps done quickly over a longer period of time), it will get you the results you want.
If you use it in anaerobically (high resistance, fewer reps, more force per step needed, done slowly), you'll build up the muscles in your legs, including the thighs. This is more like weight-lifting, done to bulk up muscles. NOT the result you want.

Beware of growing calves....when you do the stair climber, you are pushing off all of your weight from the balls of your feet, which means the muscle in your calves are taking all the strain.

setting heavy load or high resistence will increase muscle mass to be able to cope with demand, high reps and low resistance requires enduarance muscles which are smaller and will burn fat in the area thereby toning

It will increase your muscle mass there....which will in turn cause your fat there to burn off more quickly. So it's a GOOD thing.
