In gambling, it means to lose everything. The way it's used in the sense of "Chicago or Bust", it means to go all out, to do everything humanly possible to get to your destination.

Simply means you didn't make it there. Chicago ....or Bust. Your goal is to get to Chicago or "die trying". If you don't make it you are bust.

Dictionary.com give two secondary definitions that explain the use of the term:
3. Bust - Informal use
a. to burst.
b. to bankrupt; ruin financially.
In other words, if I don't get to Chicago I will burst or go bankrupt!

It means sell everything you can't take with you, and quit your job so you have nothing to go back to, therefore you have to make it to Chicago because there's no alternative.
