Truth serum is is the more common name of barbituates such as sodium pentathol or sodium amytol, and yes, they do work. Its been known to have been used in interrogations related to international intelligence operations. I'm not sure if it is still used.

Truth serum is an intravenous solution of Sodium Thiopentone ,commonly known in the medical world as Pentothal.

It is a hypnotic i.e. it induces sleep.

By constantly altering the speed ,and hence the dosage, of the I.V. solution one can get the subject in a state of partial sleep.

In that state of PARTIAL SLEEP the subject loses his inhibitions and is unimpeded by thoughts of consequences etc. He will answer questions without any inhibitions.

In this way ,TRUTH SERUM does work.

It is used by International Intelligence. I am not entirely sure whether or not it is used in enquiries of hardened criminals.

Sodium Pentathol, the original truth serum, still used today (or derivitive)for general anesthesia. Its a fine line though, to keep a person semi conscious for questioning with this drug without killing them.
