I want it to sound sincere and not cliche'. Any suggestions?

It's said all the time for a reason.Now is not the time to be clever.

Since this person is someone you know through business, I would keep the note simple, short and sincere. Perhaps you could say something like " My thoughts and prayers are with you, and and I am deeply sorry for your loss. Please, don't hesistate to call me should there be anything you need. Warmly, Brandihoo (555) 555-1234"

DId you know her brother well or their relationship?? Are you closer to her as in "friend" or "just a co-worker. If as friend, here's a suggestion:
"Jane - Words cannot begin to express how I feel about the loss of Bob. I know how close you were and I can't imagine losing a sibling so young. I want you to know you have my deepest sympathy and that I would love to take you to (coffee, lunch) sometime in the next few weeks when you feel up to it. In the meantime, if you need to talk, I'm here" - Mary
If just a co-worker keep it short: "Jane, I want you to know you have my thoughts and prayers. If you feel like you would like to talk, please do not hesitate to call me" - Mary

that is really hard, because there really isn't anything you can say to take away the pain the person is feeling for the loss of a loved one. All you can really do is show you care and that you are there for them if they need you to help them with anything, like covering for them at work, bringing them some food so they don't have to worry about things like cooking, etc.
