Its been done. I was in a show called "The Real Story Of Little Red Riding Hood", based on the book. That was a fun show.

To make the story the "little red riding hood" you first need to think about what age group you catre to, for example, if its for little kids, check out Hoodwinked and also you might want to add the word "underwear" in there somewhere because little kids will laugh at that word. For Teens, you want to have something like Simpsons humor and some crude humor(but not to crude). For adults you just need to sort of mix up the story and maybe even throw in some jokes from Hoodwinked cause even adults I know laughed at that show.

the grandma's consciousness starts speaking thru the wolf much to his dismay. Granwolf start to get into some serious personality type disorders once red arrives coz Granwolf cant decide if she/he would rather eat his/her niece of kiss her/his lunch on her cheek. Much tongue in cheek dialogue and hilarity ensues.

do it in drag...you'll blow the roof off of the place...have the guys be the female parts and the women be the mens parts...be obvious and steal some ideas from hoodwinked of course....
there is a comedy version for stage called..."Wooing wed widing hood"...the script is online....

she was going through the woods to grandmothers house n the a bear says what r u doing here, if the wolf sees u he will eat you. So she pulls out a gun n says i have this to protect me. As she finally enters grandmothers house and room, the wolf jumps out of bed and says im gonna kill you, she pulls out her gun and pionts it at the wolf, NO you are gonna EAT ME like the book says!!!!

Hmmm well it depends on the audience your catoring too, if its adults youcan try an Adult Humor(not anything pornographic) some "Family Guy" humour if you will
