I have never been to NY or even USA. but next week I need to get from 29th street to 75th quickly.
is subway the best transport to use?

Is that East or West 29th street? And is it East or West 75th Street?
Depending on which side of the island you will be, and which side you are going to, as well as the time of the day, it could take anywhere from 15 minutes to 1 hour. The subway is probably your best option but again it will depend on the time of day. If you have time on your hands take the bus, it will take forever, but you get a great view of daily NYC life.

Depending where you are on West 29th, you can walk to the 28th Street Station on 7th Avenue and take the Uptown 1 train. You will take that train 6 stops to 72nd and Broadway and then walk from 72nd to 75th.
However, if it is at night, very early in the morning (before 8AM), or on the weekend, it might be quicker to take a cab.
If you end up waiting 7 - 10 minutes for a train to show up, you could have taken a cab there in that time.

Mapquest won't help, it gives driving instructions. Take the Subway, Off the top of my head, the #1 train is your best bet - 15 minutes tops.
If it's late night, a cab might be quicker, but only if it's really late. If you do take a cab, catch one on a street heading uptown, that's save some time and money.

Subway, definitely because it would get you there the fastest. But if you have some time later, take the bus around so you can see a lot of the city.

Yes, subway is definitely best. Just make sure that you get on the local train, so that you're positive that it's going to stop where you need it to.
