It depends on what you consider losing your virginity. Some people believe its the point where you "pop the cherry" and you bleed for a second. That can be accidentally done without actual intercourse. That would probably be the medical way of losing it.
But as for the emotional and personal way, most people would say vaginal intercourse. If you've done some, but not all, you can just say you're a virgin but not completely innocent.

While you are usually considered to have your virginity until you have vaginal sex, hopefully with someone you love, it would be a good idea to be tested for STD's if you have had unprotected, risky sex with anyone, especially if you are about to make love to someone else who is sexually inexperienced and not know all the risks.

Loss of virginity is defined as "the moment penitration occurs" you can but that in your own context though be-it from a finger, tongue or penis. I define it as inserting your part into someone elses or vice versa.
