some good answers already.
Keep in mind, the discrepancies in the distances might have something to do with :
- where is the starting point and finishing point in measuring the distances? From the nearest outskirts of each city? From City Hall to City Hall? From centre of downtown?
- keep in mind, Toronto is 43 km wide, so if you start from the western border, the distance will be up to 43 km less than if you start from the eastern part of Toronto, and about 23 km less than from downtown.
- also, the distances might be different because if you measure a straight line from place to place, closest outskirts, it could be 80 km, but actually driving a car, it really is somewhat more because of curves in the roads. The way there from Toronto to Niagara is definately not a straight line. If you're driving, it is really more like + /- 130 km.
- time to travel depends on traffic and time of year. I've done it in close to 1 hour or 1 1/2 hours, but during a really nice day in summer, after the caribana festival, it took 4 1/2 hours one way because of traffic congestion. It could have taken longer, but I desperately had to find a detour, on some country roads with less traffic than the highway, otherwise it could have taken 6 hours. The traffic on the highway on that day was going at a snail's pace, all the way! There used to be a fast boat from Niagara-on-the-Lake city to Toronto. That would avoid the traffic and give you a refreshing boat ride, and great view of the skyline. I'm not sure if that is still available.

Downtown Toronto -> Downtown Niagara Falls: approx. 115km (72 miles)
Obviously, it varies depending on where you start from, and where you end.
If you're driving, the trip can be made almost exclusively on divided highways, with speed limits of 80-100km/h (50-65mph). Factor in possible traffic delays in the Toronto-Hamilton stretch, depending on when you make the drive, and you can more or less made a good guess as to the driving time.
