Champ Cars haven't run on the oval at Indianapolis since the `split' and the formation of the Indy Racing League (IRL) in 1994. So, I can say absolutely that none have run or tested on the road course used for the U.S. Grand Prix.
There is so little time between the Indianapolis 500 (the end of May, Memorial Day) and the U.S. Grand Prix, and that time is used to set up the road course. The track is then changed back to an oval format for the Brickyard 400. I'm positive an IRL car hasn't run on the road course, because of both time restrictions and because no IRL team would be willing to make the changes (to a car) necessary to run laps on the road course. They only do that for the road courses on their schedule.

No, but there have been many Grand Prix types, (now called Formula One), that have ran the 2.5 mile Indianapolis Motor Speedway oval layout. Most of these cars ran from 1909 to mid 1920's. Grand Prix cars have even Won the Indy 500, 1913 Peugeot driven by Jules Goux, 1915 Mercedes driven by Ralph DePalma, 1916 Peugeot driven by Dario Resta - a 300 mile race, 1919 Peugeot driven by Howdy Wilcox, 1922 Dusenberg driven by Jimmy Murphy - car Won the French GP, 1939 & 1940 Maserati 8CTF driven by Wilbur Shaw. The LAST F1 car to qualify & compete at the Indy 500 was a1952 Ferrari 375 driven by Alberto Ascari. The 1961 Cooper was 'based' on the F1 car, but was a 'special built' car with a 'off-set' chassis.

If by "lay out" you mean "set up" it may be possible since you set up for the track. If F1 cars compete on a track similar to Indianapolis Speedway, they might use the same set up. Of course, there are great differences in engine size, weight, aspiration, etc.

Neither an IndyCar nor a Champ Car has ever run the road course at the Speedway.
