It is not simple arithmatic gentleman. Shedding weight takes other aspects too. Discipline, diet control on continuous basis is the key. More than the run, these things are important

Once you're in shape, I believe it's reasonable to expect to lose 2-3 pounds a week as long as you stick with it. So it would realistically take you 4-6 months to drop 40 pounds. At least I believe that's the case. Remember that 1 pound of fat is 3500 calories so to lose 1 pound you have to burn 3500 calories above what you normally burn - and without eating any more calories than normal.
If you want some serious weight loss, you should exercise almost everyday and make changes to your diet as well. If you're out of shape now, don't figure you're going to be able go out and clock 3 miles right away. Start slow - maybe by taking walks for 20-30 minutes every day.
Once you finally do lose the weight, you should plan on exercising 3-4 times a week to maintain the weight and don't fall back to eating junk food.

One mile, either walking or running, equals 100 calories. One pound equals 3,500 calories. You can do the math.
It's best to do exercise in conjunction with watching your calorie intake, because you exercise three times a week but you eat at least three times a day.
However, that said, if you run 3x/week, you will start to look better faster, and you'll see results more quickly.

It depends on so many things. How much you weigh, your diet, how many calories your burning when your running, your metabolism. You need a professional to answer that question.

damn, 40 lbs, depending on what your diets consists of it could be a couple months, maybe six, though I'm not sure.
