It depends on the destination in Virginia, Amtrak's Capitol Limited travels from Chicago to Washington, D.C. A trip from Chi-town to Virginia would take about 2-3 days. A bus trip would take about 4, as for the comfort, the train would be more comfortable because you have a choice between coach and a sleeper car. The coach seats are very large and comfortable, and they lean back. Only problem with coach is the Porters and Conductors walk through the train during night stops to wake people up if their stop is coming up. A sleeper car is about the same, except the seats fold into beds at night, plus you are in your own little room. The sleeping cars' beds are made up by the porters so you dont have to mess with them. Granted the sleepers cost about 50-100 more a ticket but it is well worth it. I would prefer the train over the Bus because instead of following the long boring highways, by train you get to see things you might never see if you travel by bus.
I hope this helps answer your question, and that I didnt bore you by rambling on and on like this.

Amtrak's Capitol Limited departs Chicago Union Station at 7:05 PM daily and arrives in Washington DC approx. 17 1/2 hours later. From there, transfer to Amtrak regional service for the 2-hour trip to Richmond.

The train is a more comfortable ride, particularly if you book a sleeper for the overnight trip to DC.
