It depends on where you are.
The custom in the United States when performing Shakespeare is to perform it with an American accent. It often just sounds fake and inaccessible when the actors try to put on a British accent.

EDIT: Oh, are you talking about the word, "ay", a word that means yes? It's like "buy".

Only if you have a present day London accent would you say "Shaikespeare" (as in buy). Proper standard English and North American English is "Shaykspeare" (as in tray).
