My first answer was going to be either St. Peter's in Rome or Hagia Sophia in Istanbul. After minimal research, I have learned that Hagia Sophia is now a museum and no longer a house of worship.

I think Earl means the LDS conference center - which when it was completed a few years ago was the largest "meeting house" or it could hold the largest congregation.

But I don't think it's really the largest religious building in the world. I mean I've seen this big monastery in Portugal, and the building itself did seem quite a bit larger to me.
