According to Einstein the quickest way is not a straight line but a curve. That is because space in itself is already curved.

Depends where you are as in space you would use the gravitational pull of planets etc to increase speed so this "sling shot effect" would be quicker than a straight line journey.
Also depends on mode of transport so take your pick, there isn't any one answer to your question.

If A and B are in london I would vote for bike - beat that congestion and cancelled tubes!!

Otherwise I'm guessing the scientific route outlined by others who are cleverer than I!
