The TWAIN interface lets you acquire an image from a scanner, digital camera, or other device, and import it directly into an application (e.g., Adobe PageMaker or Adobe Photoshop). Components of the TWAIN interface are: a TWAIN-compliant application, a TWAIN Source Manager file, and a data source file for the device from which you're acquiring images.
Application
When you choose to acquire an image in a TWAIN-compliant application, it invokes the TWAIN Source Manager.
Source Manager
Applications that support the TWAIN interface provide the TWAIN Source Manager files. The Source Manager handles the communication between the application and the source device (e.g., scanner), and loads and unloads the data source. It also displays the Select Source dialog box when more than one data source is installed. When an application invokes TWAIN, the operating system loads the Source manager into memory, where it can manage multiple TWAIN-compliant applications, source devices, and data sources.
TWAIN source, file provides information about the source device (e.g., scanner) to the TWAIN-compliant application, provides the user interface for the source device in the application, and may serve as a driver for the source device. Data source files may even perform specific operations (e.g., prescan the scanner bed). While most applications use the user interface provided by the data source file, the TWAIN specification enables applications to use their own interfaces instead.

Hi,
The TWAIN standard enables multiple types of computer software programs. Today, most computer scanners include a TWAIN, TWAIN32 or compliant driver that enables the scanner to work with most computer programs automatically after they have been installed.
Complete information about TWAIN can be found at
http://www.twain.org
Your Scanner should have come with a CD which has the installation program for the scanner including the drivers and Twain 32.
You can download the required driver from:

TWAIN is the driver that is used to connect imaging hardware to your computer. So the TWAIN scoure would be the scanner. It's saying it's not finding the scanner. And it needs a driver to find the scanner. When you said you downloaded the software, did you also download the driver? The software is just the application that allows you to manage your scans. The driver is what enables your computer to talk to the scanner. Download and install both the software and driver. I'm not familiar with the software, but it may also require some setup, specificially, you have to select the scanner, even if the scanner is the only TWAIN device, it may still require you to manually select it.
