I'm at the grocery store - could be that you just drove up and are in the parking lot.
in sounds better to me...

The only difference between the two is the implication of your location. So... are you in the grocery store or at the grocery store?

Both work. I would say either if I was in the store. However I wouldnt say "Im in the Grocery Store" if I was outside.

Depends on whether you are in front of the grocery store or inside it. If you are on your cell phone in your car in the parking lot, then #1 is more correct. If you are on your cell phone at the Deli counter, then #2 is better.
