It depends on what your interests are. Central Park is a great place and best of all, free! There are many different parts of the park to enjoy. There is a fee if you want to go to the zoo, but it's a small zoo, so I myself wouldn't bother.
If you like history, the Ellis Island is a MUST! Even moreso, IMHO, than seeing the Statue of Liberty, which while impressive and interesting is not the real story. The real story of America and immigration lives at Ellis Island.
If you like Museums, the The Metropolitan Museum of Art (on Fifth Avene in Central Park) is a MUST! They have art from ancient times to current day. There is a $20 suggested donation, but if you are a student it's only $10. You can spend an entire day in the Museum and not walk through the same hallway twice.
I'd also suggest that you go to a local bookstore before you come to visit and look at some guide books. Perhaps you will find one that appeals to you and what you enjoy doing. It would be a worthwhile purchase to study up a bit before you get to NY. It's really an amazing city with so much energy to spare. Yes, it's dirty and it's noisy, but it's the heartbeat of America!

Empire State Building and/or Top of Rock
Ellis Island
Ground Zero
South Street Seaport
The Cloisters
The Guggenheim, the Museum of Modern Art, the Metropolitan, and the Museum of Natural History
Radio City Music Hall and Lincoln Center
A couple of Broadway/off-Broadway shows
St. Patrick's Cathedral and the Cathedral of St. John the Divine
Tiffany's and the Trump Tower (window shopping only)
A bus tour will provide you with a good initial overview
.... This doesn't begin to scratch the surface. The "best" place will depend largely on what sorts of things interest you.

Many people try the Empire state building. But, that's not the best view of the city. Try Rockefeller tower ("Top of the rock") and they even provide a nice pamphlet to identify a bunch of buildings on the NYC skyline, day or night. If you have a lot of time, reserve a seat at a show in the Radio City Music Hall. I saw the "Christmas extravaganza" and it's definitely worth your money.
