By 'best', what do you mean? Most comfortable airlines to sit in? Location on an aircraft that gives you least airsickness? Least noise? More leg-room? Seat that gives you the best view out the window?
Over the wings or in front of the wings will give you least feeling of airsick - this is why First Class is at the front. In fact, inside the cockpit you can't feel anything, even when turning. Behind the wings could make you airsick if you're prone to this, and the further back you sit, the more pronounced this feels. But, the further back you sit, the better the views, as the wings and engines don't obstruct the view out the windows.
Sitting at the emergency exits (the Overwings) will give you a lot more leg room. However, you must be a physically fit adult to sit there, as you may be required to open the exit in an emergency.
For less noise, the front is best. Aircraft are noisiest from the back of the engines, where the thrust comes out. In front of the engines, the noise is slightly deeper in tone, not a high-pitched scream. Once again, this is why First Class is at the front.
As for most comfy seats .... in Economy Class, most airlines have seats that all feel the same. Its in Business and First Class that airlines differ. But no mere mortal can afford the fares, so it doesn't matter to 99% of the population.

Obviously the best seats are in the first class section, but in the rest of the airplane the seats with the most leg room are in the rows that contain the emergency exits. But if you are looking for comfort, chartering a private jet is the way to go. Here is a site that shows you private jets selctions.

I have travelled on many aircraft...but I have never sat in the cockpit.
Best to ask the pilots.
