Manhattan has the fortunate advantage of being very compact, so all the big landmarks are pretty close together.
I would recommend the Gray Line open bus tours, as they are relatively cheap and show you all the main attractions.
A trip up the Empire State Building is a must too - the view, oh my god!
Staten Island isn't that good from my experience, but I may have gone on a bad day.
Central Park is very nice, especially at this time of year, very romantic.
Bloomingdales is just like Debenhams but bigger, and the Rockafeller Centre Ice rink is tiny! But when it's all lit up, it looks lovely.
Have a meal in China town - it'll be the best Chinese you'll ever have!!

Empire State Building (for the observatory),
SoHo,
ChinaTown,
The Port
Statue of Liberty
The WTC site
5th Ave/Madison Ave for shopping
Maybe see a Broadway Play or off Broadway Play.

St Mark's Place, Greenwich Village, lower Broadway near Chinatown are probably the best for 20/25 year olds- that's where we always go. There are awesome stores (H&M, Sephora, YellowRat Bastard, Urban Outfitters, Trash and Vaudeville, etc). That part of Broadway also has this awesome Mexican bar called Gonzalez y Gonzalez that doesn't card too much and have a great happy hour.

There is plenty of sightseeing to be done in New York such as the Empire State Bldg, Times sq, China town Central Station etc. if you or your girlfriend like shopping you can try Macy's or Bloomingdales as well as Tiffany &Co. Try the ice skating in Central park too!
