The best way is to see how they respond to a compliment... I think you know the difference between how a straight girl would react ("oh thank you, you think???") and a lez or bi, who will tilt their head and smile graciously.
Then jokingly say "if I my name was George, I would ...." At which point they will laugh, and then you say "Or if you weren't straight, that could work, too" After which they should disclose whether they are or not.
Mostly just be friendly, have fun, look them in the eye and TILT YOUR HEAD, you have no idea how much this one works.

It makes it much easier if you have a group of GLBT friends. I would suggest going to events, clubs and things that you know more gay/bi women are likely to be at. Find a good lesbian friend or bi friend to hang out with. I have also had a lot of luck online with myspace and things like that when meeting women.
There will be straight girls who come on to you and want to experiment with you. You need to set clear boundaries with friends, especially when drinking. Most friends are able to be mature enough, however there are some instances where good friendships can be lost over such things.

Ask them to meet you at a poetry reading or ask if they can help you hang some pictures in your apartment. They like peace and guitars. I'm straight by the way

Well, perhaps you should just imagine them as another guy! You know...it just helps when you feel like there's no pressure that way...and if nothing happens, it's ok...you'll just have more experience talking with someone you find attractive!

I'm also bi-curious. I would suggest going to lesbian clubs - that way you know what the women are looking for and it saves the trouble of having to ask or wonder. :)
