As has already been said; if he's changed his mannerisms or appearence for the better... maybe changed jobs or social circles recently. Has he been spending less time with you, or changed at all? Does he seem stand-offish or secretive?
Think about why you are suspicious - are you justified? Has he cheated or show signs of cheating before? Is there anyone else close to him you could confide in, who might know or notice similar differences in him?
Ask what's new with him; try to get involved more... if he is cheating, he will probably resist your increased involvement in his life, and you'll have your answer.

afta? By the way he acts. When he starts wearing his best clothes, when his grooming suddenly changes. For instance if he hardly ever combs his hair, then one day out of the blue he has someplace to go, and he gets all dolled up just for this outing. He takes a shower, shaves, combs his hair, brushes teeth, splashes on cologne and puts on his good clothes. That right there should be a hint that something is not right.

when he acts strange/different around you.
usually the gf/wife begins to suspect things, i'd go with instinct.
but make sure you have proof that he is or else you'll never get him to admit it.
