well...........
in the first case.............it is correct.....because verb To Be dont need auxiliary verb

the second one, if you'll analyze it grammaticaly, is not correct.

1... DO in that case is the auxiliary verb..... so.......which is the verb that DO is the auxiliar to???

You always have to use Do you.......... at home?

For example.....Do you cook at home?
Do you study at home?
Do you sleep at home?

May be some pple can use it as you wrote , but is not correct.......at least in the correct english.

1 is good, or you could just say "Are you home?"

2 could be contrived to have a meaning, although most likely you'd need an apostrophe: Do you at-home? Meaning, say, do you work at home, or are you a stay-at-home parent, or do you homeschool, or do you stay home during your vacation? It doesn't mean "are you in your own house right now?" though.

the seccond is really bad english.
the first one is not wrong.
However, "home" is a special word that does not require the preposintion in phrases like "go to", "stay/be at".

So we say:
"I'm going home" not "i'm going to home";
"i'm staying home" not "im staying at home";

And:
"Are you home?" this is more natural sounding than "Are you at home?"

You're kidding me? "Do" is just an auxiliary verb, here, it really needs a main verb to make a complete sentence: "Do + Subject + main Verb" = Do you learn?
The first one is correct.

"Are you at home?" is an acceptable question. If however you want to ask something somebody does at home you should ask: "Do you, at home...." followed by your question
