With a knee injury, the exercised should be monitored by doctor. To take off 40 lbs, also should be monitored by your doctor. Taking all kinds of advice can cause you more injury to your knee and it is best to do what the doctor says.

Pamper that knee because it is very vulnerable. Your elbows and knees are the weakest joints in your body. But you don't support your weight with your elbows.
Start with walking. I would recommend wrapping your knee in an ace bandage for support. Walk until you begin to feel something in your knee and stop. Elevate that leg slightly afterwards. Slowly increase your time/distance. Be patient. If you rush it, your knee will scream with a reminder that it's still vulnerable. Good luck!

Pilates! It was actually invented for people who underwent an injury :)
However if you want to lose weight (not just exercise) you need to change the way you eat.
