I always throw my coin in The Fountain of Trevi. That way I'm sure I'll be going back.

An impossible question!

I would have to answer - go for a walk. It's amazing what you can find and many of the big sights are in walking distance of each other - Spanish Steps, Trevi Fountain, Pantheon etc

I would also maybe say go to the Pantheon (or anywhere near a major tourist attraction), sit down at one of the cafes and just have a nice coffee and watch the world go by! People watching is fascinating.

Mostly, it depends what you are into -
For art or religion go to the Vatican & Vatican Museums and see the Sistine Chapel.
For history see the Colleseum and surrounding area.
For food go and eat in Travestere.
For people and architecture- everywhere!

Just mind your pockets and bags and take loads of money - Rome is bloody expensive!

I agree with the answerer who said it's impossible. But since you press me :) for just one:
I'd pick a Wednesday: get a one-day free pass to inside the Vatican city (not needed for St. Peter's or St.Peter's Square), stay on the square first, take part in the Pope's General Audience, then use my pass to go to the Vatican Gardens. On the way back I'd enter the Basilica.
Oh, that was 3 things. Whatever...

Eat gelato. Seriously. Its the best thing to do over in Rome. In my opinion, Rome is a bit overrated. So if you can only do one thing, just go and eat some gelato. =)

tossup between the Vatican, visiting the Pantheon, and visiting the Coliseum...couldn't go wrong with any of those 3.
