When I sit or lie down, I have no pain or problems at all. It is so severe now that I can't do anything that requires standing or walking for more than 5 - 10 minutes. I am a 40 year old female, and I have gained some weight in the past few years. I have never had back sugery or any specific injury.

Yes weight can be a factor but not all the time. It could be spasms, which is what you describe as a "knot". Are you exercising in any way? some light exercise can help you, or stretching can help too. First of all if the pain is too much, please see a Dr. i can be something more serious than a muscle spasm or trigger points.
Heres a site where it shows you some stretches for your lower back
I hope you feel better
Good luck ^i^♥

From experience it's the excess weight. Try to lose the extra pounds and see your doctor for some advice on pain management and walk as much as you can.
