Of course there is, but it will be a very long, long ride. The flight distance is about 1500 miles that takes you straight across the Mediterranean, but by land, you will have to go north to get to the Riviera, then travel east through France and Italy into the Balkans and eventually pass through Bulgaria or Greece to enter Turkey west of Istanbul.
I once took the train from Istanbul to Budapest through Bucharest--a trip that was nowhere near as long as the one you are thinking about--but what a trip it was! I can't imagine that the bus can be any worse and may be better. But I'm just guessing that it will take the better part of three days and nights.
However, I'm also thinking that it will be an adventure that you will not soon forget. If you do this, you will travel roads that were the trails of invaders and conquerors over the past 3,000 years or so, as the site of Istanbul was the one place where great armies could cross easily from Asia into Europe. Get a good historical guide book to read along the way.

If you are decided to do that there is a way.
www.eurolines.com partner companies can give you a connection.
You can take the Alsa bus from Madrid to France, Lyon and then take an Ulusoy bus from Lyon to Istanbul.
Other options are:

via Munich Germany

I m not sure about a bus from Madrid, but there is bus service from Munich to Istanbul everyday.

well. u'll have to cross the Mediterranean sea.. so i don't think can cross it with a bus... maybe u can take the but 2 a port and after that a boat........ forget about the bus.. u should take a plane..
