If you are single and a woman, I would go with inside entrance and other than ground floor room. Safety is priority. Location in the area is next...you don't want to be out in nowhere and have no place to eat.
Room service is a great amenity if you are working and don't want to go out. I like to have room service, so I can relax, do internet, eat and watch television where I am most comfortable.
If you are going to entertain or do business, a suite is great as it kind of seperates and lets you have some space to yourself.

It depends on how long you are at that hotel.
As long as its got the usual bed/shower/toilet/tv I'm fine if its for a day.
More than a day, I'd like it to have a fitness center, free internet access, possibly an indoor pool, maybe a jacuzzi in the room, continental breakfast. Fridge and microwave would be nice too.
And hopefull peace and quiet :)

It has to be the Maid service, I always look for a clean room it doesn't have to be so Fancy as long as its clean, I was in a hotel in Montreal and boy it was so great the beds were all in white linen and even the beautifully comforter cover was white as white can be and soft and comfy beds. guaranteed my repeat business time and time again.

fridge(even a bar fridge for water etc)
extra linen & bath towels (you can never have enough)
room service
24hr reception is a nice to have - coz you can call them for anything you may need
then the other nice to have' pool, sauna, gym

A good and clean bathroom, clean sheets, a cafeteria, silence. At least, this is a minimum.
