How do they affect the audio quality and what's the best setting?

44.1 kHz is the sample rate you should use it coincides with the sample rate of an audio cd. As for the bit depth that is the amount of compression. MP3 being a lossy format the more compression the more quality is lost. The most acceptable bit depth right now is 128k however 192k is becoming more popular. Also the lower the compression obviously the file size will get larger.

128k X 5 Minutes = 4.6875 megabytes file size
192k X 5 Minutes = 7.03125 megabytes file size

Higher sample rate the better the reproduction, but also the larger the file size. Never heard of bit DEPTH in audio. That seems more of a video monotor parameter. There is a bit RATE and again the higher the better
iTunes has a pull-down menu you can use, I just max it out.

It wont be any more beneficial, no matter if it says MP3-320 Kbps. you may upconvert a record, you may basically downconvert it. Going with a 320 Kbps VBR bitrate provides you you with large high quality on condition that you're ripping off a disk (that is a very severe lossless record format, considering, on widely used, you've 11 songs on a 700MB disk thats round 60MB a music).
