It depends what sort of stuff you're into. If you head into the north, you can check out the Great Barrier Reef, although I think it rains too much. There is also Fraser Island off of Hervey Bay and the Whitsundays off of Airlie Beach, although both towns are just springboards for those off-coast destinations. Watch out though, because some places in Queensland get lost of rain, and some prt of the coast has box jellyfish which means that you may not even be able to swim. Melbourne is a great place though, as is Sydney. You can spend a few days in each, and each city has a worthwhile daytrip, especially Melbourne (Philip Island, Great Ocean Road, etc.). Loneleyplanet.com has a brief summary of some highlights in each country, and I figure that holds up for Australia as well.

Why limit yourself to cities ?
Australian cities in any event are possibly quite small to someone used to a real-sized city. Don't forget that there are only 20million of us in the whole country.
Australia is a vast island continent and has so much to see and do, so it depends on what your timescale is and your mode of travel.
If you wanted to take 3 or more months, fly into the top end (Darwin) hire or buy a campervan, head through Kakadu National Park, visit the Great Barrier Reef, head down the Queensland coast , visit Brisbane, Sydney, Melbourne, Adelaide and then drive across the Nullabor to Perth.
Then head up to Shark Bay to visit the dolphins, carry on North up through the Pilbarra and Kimberly and finally back into Darwin. .
Then fly back to Singapore.
I see that I've missed Ayers Rock and Alice Springs in this, so you'll have to review the route once you realise how much there is in Australia
