Oh my god, do not go to Times Square, or Herald Square, or any "square" period. They are tourist traps and not like the rest of NYC at all. I am so tired of people visiting NYC but only going to tourist places in Midtown and then saying they have no idea how anyone could live here. Well, no one in NYC lives in that area, that's why. I go there maybe twice a year, and only if I'm trying to go somewhere else...
Go to the West Village. It's so pretty... cobblestone streets, adorable little apartments, great restaurants and cute little bookstores and coffee shops and oh, if I could afford it, I'd live there in a second. Take the blue subway line or the orange subway line to West 4th street or the yellow line to 8th street. (Oh, and take the subway, not a cab, at least once. The subway rules.)
Go to St. Marks for record shopping, bars, vintage stores, sushi, etc. Green line to Astor Place.
Go to the Lower East Side and just walk around. It's a fun place, period. Take the F to second avenue.
Finally, go to BROOKLYN! We do have other places to see besides Manhattan. Williamsburg is really cool. And I live there. But I'm biased. Take the L to Bedford ave. Oh, and take the Q to Beverly Road or Avenue H to see some really cool victorian houses.
Just please... don't go to Times Square! Or if you do, go other places too.

It's hard to answer because I don't know you and your three friends interest since New York City has a LOT to do. You can't go wrong with checking out Times Square or perhaps do something touristy like checking out the Empire State Building. At night...TONS of nightlife activities...clubs, bars, etc.

NYC ~ Empire state building and statue of liberty are a must...I've also heard that the 9/11 memorial is worth a visit, but I've never had the opportunity to go.
NY state ~ Niagara Falls, Howe's canverns, Lake Erie, Amish country....
