First, exercise without changing diet will help you lose weight. Changing your eating habits will help accelerate the process.
I'd highly recommend a book called 'fit for life' to help plan your eating.
But for the treadmill, the idea is to get on and just START MOVING!! You don't want to run, but start off with a light walk for 15 minutes the first day. Increase the time on the treadmill by 2 minutes each day until you reach about 45 minutes - 1 hour. Yeah, you need to be doing this for 4 or 5 days a week.
As you get your cardio system going, the goal is to warm up for 10-15 minutes of nice, easy walking. Then, hit a 'plateau' where you'll be burning the carbs for about 30 minutes.
THIS IS NOT RUNNING! You want a brisk walk, where you could just be holding a conversation with someone, but not huffing and gasping for breath. If you can't talk and do this, you're going too fast.
Then, a 10-15 minute cooldown. DO NOT SKIP THIS STEP!!! ;)
Finally, I highly recommend you do this in the morning. It'll wake you up, get your metabolism cranked for the rest of the day, and after a couple of weeks you'll be noticing your energy is skyrocketing.
Good luck!

You will get the best results by using it regularly. That's more than most people can manage.

start slow and increase the speed as you go. Then slow it down before you finish. You will loose TONS of weight (figuratively speaking).
