I'm not sure just what you mean about the one thing?
I believe that Judas Iscariot knew exactly what was to happen when God used him to be the betrayer of Jesus Christ.
Unlike today, people then knew their Scriptures. They were waiting for the Christ, the Messiah to arrive. They knew what was the role of their Messiah was.
At the Last Supper, Jesus turned to Judas Iscariot and said, go quickly and do that what you must. (paraphrased)
Can you imagine in the first earth age after the attempted overthrow by Lucifer, what the souls that sided with God were going to have to do? I think Judas was right there with them. God knew he had the strenght to fulfill this duty and Judas gladly accepted it.

That the death of Christ would put into effect the entire atonement and ultimate resurrection from the dead. What Judas thought would be a negative thing (Christ's death) actually resulted in the salvation of all mankind.

There was alot he didn't know and not just him all of his disciples. I'm not so sure if there is the "One" thing he didn't know except maybe he was God in the flesh and had the situation in his pocket...
