I've used Seroquel both for long term treatment as well as a 'prn' medication, meaning an extra smaller dose if I'm really wound up or agitated.
The PRNs usually work within a short period of time. If you are using Seroquel for schizophrenia or mania, it may take several doses over a period of days or weeks to become fully effective.
Unfortunately, almost all antipsychotics have weight gain as a side effect. That doesn't necessarily mean you will gain weight, though.

I was on seroquel for about 6 years. I was prescribed it when i was 11 years old and stopped using it when i was about 17.
I gained a LOT of weight on it. I gained about 75lbs (over the course of those 6 years.) It has been 2 years since I have taken it and I am still have about 45lbs to loose until I'm at my normal weight. ( They had me on 200mg a day.... and I was 11 years old....my mother is 39 years old and they have her on 25mg)
I dont feel that it helped my mood, other then knocking me out and giving me a good nights sleep.
My mother recently stopped taking seroquel becuase of the weight gain....she was only on 25mg so the weight gain wasnt very noticable to her at first...
Now she is on Effexor. I was also on it and it worked wonders for my mood (as well as hers). It began working I feel within the first few days of taking it.
My mother still takes effexor and I notice that she is a much more stable person than she used to be. .... (I did not stop taking effexor for any reason other than it gave me very bad headachs and I'm doing okay without anything).
You may get different results with the seroquel. All of our bodies are different. Dont give up on it just yet.

It's fairly fast acting...took a few days though. Caused HUGE weight gain. Only been on it 3 month and have gained 30 lbs. It makes you hungry all the time. Even after you have just finished a meal your brain never tells you that you are full. It is so strange. Told doctor today I would not take it another day after reading extensive info on internet concerning class action lawsuits for it causing diabetes in patients. They are trying to recall it. I think there must be another alternative out there.
