There are two locations for TKTS - Times Square and South Street - read the locations and hours here:

Take cash - they don't accept credit cards.

Write down your top five or six choices before you get there. Some shows never show up at TKTS - Wicked tickets are not available - last week they did have Spamalot, Chicago, Hairspray, etc. You might also consider Off-Broadway - some of the shows are great and usually cost less than half the price.

If you are worried about your seats, you can purchase a booklet called "Stubs" that has a seating plan for every Broadway theatre. There are very few really bad seats, though. Once, I had one of these losers - there was a big pillar right in front of me and I had to sit at the edge of my seat and crane my neck in order to see the stage. That doesn't happen often.

If you see empty seats that look better, run for them at the intermission.

Where you sit does matter especially if you are way to the side in front. I did once and could not even see part of the stage. The cheapest tickets are at TKTS in Times Square the day of the performance.

Tkts is fine if you want to waste hours in line.Best to get tickets on-line (you can print them out and bring to theater), we got tickets to Drowsy Chaperone (very good show) that way, we had the cheapest tickets (less than $30) and they were fine, most theaters on Broadway aren't that big.
