Yep. Whether it's medication, food, a soap, whatever - if you're constantly exposed to a substance, it's not at all unusual to start reacting negatively to it.

as you take the drug in the first time your body become sensitized (makes antibodies against the drug) if you are genetically predisposed, however there are no symptoms of allergy as the antibodies are being made and not enough to induce allergic reaction.
in the second time (the body is sensitized and antibodies are now made and enough) you take the drug (regardless the time interval between 1st and 2nd exposure after the sensitzation process is complete), it will react with the already made antibodies and induce allergic reaction.
sensitization process usually takes 10-14 days average which there are no symptoms ,but it varies among people.

Yes...if I'm understanding the question right, you can suddenly develop an allergy to a drug you've been taking for awhile.

Yes you can. If you fear you are having an allergic reaction to your medications, you should discuss it with your doctor right away. Your doctor will weigh the allergic reaction to the benefits of the drug. S/he may decide to keep you on the same prescription or could change you to another.
