The setting (time and place) influences characters by changing the environment from which they came. A character's personality is defined by how he interacts with other characters and the environment within the story. For example, a female character who is from the deep south in the 1800's is expected to be a "proper" Southern woman. It will affect her speech, actions, and attitude. In contrast, a character who lives in the slums of Chicago in the 1960's has an entirely different upbringing and outlook on life.
