Build yourself meaning with your own hands or designing it and hiring the sub-contractors to do the labor. I would recommend the later. Even with custom built homes, there are so many decisions to make. You even have to tell them what hinges, nails, thin-set, size wiring, everything you want. Let the professionals do as much as they can /know. Unless you really have the necessary skills to do so yourself. Even with your plan to educate yourself on all the trades, it will take many, many ( at least 10) years of education.

In some cases - yes. It depends on the cost of the land and builder rates in your area. I would recommend a modular home if you want to go that route. Modulars are not like trailers or manufactured homes, they are real houses that can be designed in any style and are built in factories, shipped to the location and assembled by an experienced contractor. Panelized homes are another option, but they are not complete like modulars and require more labor to construct. Many builders of popular homes like Ryan, Toll Brothers and K. Hovnanian use a combination of modular and panelized construction. I live in a townhouse community and all of the homes are panelized. Look up the terms "modular house" and panelized house" on Yahoo and you'll see what I mean. Barnes & Noble also has good books on both subjects. Penn Lyon is a good modular builder and Eagles Nest Homes is a good panelized builder.

By build yourself, do you mean the carpentry and electrical and the concrete ? OR do you mean you will be the general contractor who manages the subs ?
If you mean literally do all the trade work yourself, then you might finish by the time you are 40 yrs old OR
if you mean be the general contractor - that is do able,
go for that .

YOu need to obtain the proper licences for various tasks in order to get the permits to do the building.
