No. Different pigments cannot be detected in spinach before paper chromatography.

The dominant pigment , Chlorophyll, always masks the other pigment.

That is precisely why the chromatography is performed.

Due to their difference in the rate of their movement ,they separate independently along the strip of the paper . Thus even the tiniest amount of pigment can be detected.
