There was a song in the late 50's early 60's era. The girl singing was asking her boyfriend for his comb. She called him Kookie.
My guess from that is it is a nickname

Either it means cookie like the tasty dessert or it means the other way which is crazy. If someone calls you kookie they either are trying to say you are weird or you are sweet like a cookie.
