I am a 16 year old boy. When is the best time to do it and what should I do?

Depends what you want to do. I am guessing though that you are interested in building muscle. For the first few weeks just go in 4 times a week doing everything you possibly can. Circuit train. do your chest, traps, delts, bi's, tris, abs, everything Then once you have killed yourself you will have found that you have built up considerable stamina. In these first few weeks do not go in with any expectations. You will lift what you are able to--don't strain yourself.
Now is the time to start regimenting your workout. I recommend doing two different muscle groups over a course of 4 days in a week with at least one day also devoted to cardio. Day one can include chest and shoulders. Day two is legs, ***, and abs, day three is arms, and day four is chest and shoulders again with abs. Then pick up a cardio regime. Sweet, I'm 17 so I know what your goin through.
Start off pretty slow, don't overdo it. If you are doing weight training, gradually increase reps (8-12) and then the weight. For example, bench 50 lbs 8 times, then 10, then 12 when you are ready, then increase weight. If you are looking for cardio, running is the only thing I can suggest.

Now is the best time, set time every other evening to workout.
Get u a set of weights if possible.
Start out by doing stretching, do side stretches,arm stretches,etc.
Then do situps, weight lifts,etc
