If you are referring to Tylenol and aspirin and ibuprofen then you need to be careful about the amount and the time frame in which you take them. For all medications always follow the directions, either the doctors instructions or whatever it says on the bottle. If you take to much of the over the counter ones they can shut down your kidneys which is a very bad thing. They can also damage you liver. The side effects depend on what pain killer you are referring to.

Do not ever take more than is recommended on the bottle unless directed to by a doctor. It will tear up your stomach and some of them like Tylenol have adverse effects if you take too much for too long and then try to stop taking it.

pain killers can do a number on your stomach, that's the reason why you are supposed to take them with food.
