The Chinatown bus runs from Chinatown in Philly to Chinatown in NYC. It's about $12 one way, $20 round trip right now. It's the cheapest/easiest way to travel between the two cities. It's a 2 1/2 hour trip and the buses leave every hour (I think) I've done it before. The bus seats are pretty uncomfortable but it's cheap and basically hassle free. This link has all the info:

If you have a car, I guess it takes about 2 hours, give or take. It would be easiest late at night when there's less traffic. The bus is better since you don't have to worry about parking or parking tickets, etc..

The Amtrak Acela Express goes from NY Penn Station (33rd Street Between 7th & 8th) and 30th Street Station (30th and Market) in as little as 1:08.
