Before the war began, Lee was a Lt. Colonel in the U.S 2nd Cavalry. After Virginia seceded from the Union he resigned his commission and returned to his native state and at the outbreak of the war he was given command of all Virginia's forces. When Jefferson Davis formed the Confederate States Forces, Lee was named one of its first five full generals. In June 1862 after General Joseph Johnson was wounded, he was appointed Commander of the Army of Northern Virginia, this was his first opportunity to lead an Army in the field.
Lee never wore the rank of Confederate General. Instead he wore the 3 stars of the rank of Confederate Colonel, equivalent his last U.S Army rank.

He played a huge role. He was the Primary General of the Confederacy. When he surrendered at Appomattox Courthouse in 1865, it caused many other Southern Units still fighting to realize that they were defeated and surrender to the North, bringing an end to the Civil War.

He was commander of the rebel forces. He was well liked and respected even after the civil war ended. He was defeated but the union army was much larger and had better resources.

Get a history book focusing on the Civil War.
Step two:
Read it.
Seriously, people spend years studying that war and still ask questions about it. I am afraid any sort of a meaningful answer here would be impossible.
