There are a number of ways...
1: Some states have a Catalog of Philanthropy that highlight worthy charities each year. You can check it out on-line.
2: Think of organizations that have meaning to you - the school or camp that you attended that impacted your life, if someone in your family has been effected with a disease or disability, give to an organization that provides services or conducts research.
3:Look in your local community... there are so many organizations doing great things that desperately needs support.

Find what is right for you.

I see that there are a number of good suggestions and information that have been offered to you already. Perhaps you can take some of this information, decide what criteria is important to you, and then do a search based on that criteria.

There are two tools in particular that I would suggestion:

www.charitynavigator.org

www.guidestar.org

These sites will allow you to get additional information about the mission of various nonprofit organizations, various ratings about the fiscal efficiency of organizations, etc.

Or, if you decide you don't want to just do a strict donation, there are several ways you can support charitable causes through purchases. One is through eBay's Giving Works program (givingworks.ebay.com). Each listing through this program will state the donation percentage (that is, the percentage of the final sale price that will be donated). The program is powered by MissionFish, who verifies organizations as well as ensures the funds reach them.

Obviously, I think this is a personal decision. You should give to who and whatever you feel is most needy and deserving. My best advise would be not to get in a routine of giving the same amount to the same charity all the time. Pay attention to needs in your community. There may be families in your area you could really help that have a temporary need. (for instance medical needs, house burned, etc.) And if you want to keep the whole giving thing from being awkward for you and them both, you can ask a local church to give your donation to them in the churches name. Lots of times people will accept help from a church when they wouldn't take help from an individual.

Look for a charity that has a history of impact in the community without a majority of funds being spent on "overhead" or the workers in that charity.

either give it all to one like a childrens hospital, or split it up between heart, lung, kidney, breast cancer awareness, or give it to a homeless shelter, food bank, or your local charities. I always help my people locally, that way i know that the money is helping the people in our community. I donated a lot to the kids in our community that don't have toys for christmas.

Given the huge variety of need, there is not really a best but what would inspire you ?
Red Cross helps people with immediate disasters . . .
Salvation Army helps people with substance abuse issues get back on track with long term programs . . .
And a number of programs in Oprah's Angel Network deal with youth education.
Maybe find 2 worthy ones and give each $500 a month.

Whatever you feel strongly about.
I tend to lean toward children and paralyzed organizations. I had 2 family members who were paralyzed, and I think Make a Wish is nice for kids.
I have a soft spot for older folks too.
Any will do it is just the giving that counts!

The best is an auction on a mystery box auction on ebay. I currently have oe running for Toys for Tots. The reason these are the best is because the money goes to charity, and you get Awesome holiday gifts for yourself and family for donating the most to the auction. Visit the auction and check it out!
