We are going down to a Knicks game from upstate NY and staying overnight, want to bring a small bag, change of clothes, etc. and we know we cant bring it into the game, and are looking for a locker until after the game and drinks

There are no public lockers at Penn Station.
What you might want to do is find out if Play by Play is open before the game. That is a sports bar/restaurant that is on the club level. They have a coat check there, and would probably be able to take your bag.
It is usually only open to club members, but you might be able to make a reservation.

No, there used to be but after the 1st bombing of the world trade center they took them away. Just not safe anymore :( The whole coat check idea is a good one. Or just stop back at your hotel and change.
