Where: at the gate? at check-in?
When: early? 5 minutes before boarding?

(I'm taking an international flight, don't have elite status :/, and would maim someone for an aisle seat.)

Checking in online sometimes is not in real time like it is when you call the airlines. You see, 24 hours before your flight, call up the airline. They will release all of the elite status, exit rows, and handicapped seats in the bulhead. Be the first one to call so you can get one. People call reservations all the time saying, they have had knee surgery, bladder problems etc just to get an aisle seat. If you do have one of these problems, make sure you call the airlines and tell them you would like to submit a "service request" for an aisle seat due to "whatever". They will notate it for the Airport Gate Agent to see.

Tell your travel agent when you book the flight that you have
a bladder problem and must sit in an aisle seat. Need to get to
the bathroom fast.! I am sure the other passengers would agree.

If you go to the airlines web page you can pull up a seating chart and change your seat, even before check in. If there are blocked seats, which airlines keep for people with frequent flyer status, you can obtain those seats, if empty when you check in for your flight.

call the airline a few days before your flight. They'll be really helpful. Tell them you want an aisle seat and they'll do their best.

ask the check-in person as soon as you check in. if that doesn't work, ask the flight attendant as soon as you broad, if that doesn't work ask a person who seems to be traveling alone.
