Depends where on Long Island. JFK is in Queens which technically is Long Island. Note that the boroughs of Queens and Brooklyn are both physically on Long Island. Nassau County is where the Long Island region begins. With no traffic, this is 10 minutes from JFK. Montauk Point which is the end of the "Long Island" Region so to speak is about 3 hours away.

JFK is in the southern part of Queens, part of the inlet area called Jamaica Bay. To the direct east of the airport is Nassau County LI, and the island extends another couple hours east of that point out to the Hamptons. So to answer your question, it should take only a few minutes to get from JFK to the western extreme of Long Island. In fact, the airport is closer to LI than it is to Manhattan.

Long Island CITY is in Queens, and is probably a 30 minute drive to JFK.
