Depends on who you're talking to. The verb 'to know' is Saber.
If your talking to a family member or a close friend, you use the informal second person singular 'sabes'. If you're talking to someone you don't know, espically someone with senority over you, use the third person, which is also the second person formal 'sabe'. You could put tu (you, informal) or usted (you, formal) in front, but it's not common for spanish speaking. So:
?(Tu) Sabes mucho?
or
?(Usted) Sabe mucho?

In Spanish there really is not equivalent to change something from statement to question form. From "you know a lot" to "Do you know a lot?" In Spanish, "sabes mucho" to "sabes mucho?", it is only really the way you pronounce it.

You know a lot = tú sabes mucho
Do you know a lot? = ¿Sabes mucho?

You know spanish = tú sabes español
Do you know spanish? = ¿Sabes español?

¿Sabes mucho? -Informal way

¿Sabe (Usted) mucho? -Formal way

Use the second way to talk to people you don't know, elders, teachers.....etc.

Use the first way for people that you know (family, friends, children, etc.)
